Workflow Process

Staining

 Flooding the bacterial smear with crystal violet, a purple dye to stain all cells on the slide

Decolorization

Rinsing the slide with ethanol. This step differentiates between GRAMPositive and GRAM-Negative.

Counter Staining

 GRAM-Negative will take up the counterstain and appear red under the microscope.

Washing & Drying

Rinsing the slide with water to stop the staining process and drying the slide.

Microscopy

 Observing the stained bacteria under a microscope.
